Single Molecule Imaging by 2-Photon Excitation — •M. Sonnleitner, G.S. Harms, G.J. Schütz, and Th. Schmidt — Institute of Biophysics, University of Linz, 4040 Linz, Austria

Wide-field two-photon excitation was exploited to study the diffusion of individual fluorescence-labeled lipids in a supported phospholipid membrane on a millisecond time scale. A CCD-camera was used for direct imaging of individual molecules.

The two-photon approach allowed for low background parallel observation up to a field-of-view of 5µ m size. The signal-to-noise ratios are compared to conventional one-photon measurements in terms of excitation wavelength, laser intensity, and illumination time.
