Wide-field two-photon excitation was exploited to study the diffusion of individual fluorescence-labeled lipids in a supported phospholipid membrane on a millisecond time scale. A CCD-camera was used for direct imaging of individual molecules.
The two-photon approach allowed for low background parallel observation up to a field-of-view of 5µ m size. The signal-to-noise ratios are compared to conventional one-photon measurements in terms of excitation wavelength, laser intensity, and illumination time.
