Simulation of Iodine-125 Induced DNA Strand Breaks within a CAP-DNA Complex — •Wei Bo Li, Werner Friedland, Peter Jacob, and Herwig G. Paretzke — GSF-Institut für Strahlenschutz, 85764 Neuherberg
The yield and distribution of strand breaks induced by iodine-125 in individual nucleotides of the DNA in a CAP-DNA complex has been simulated in the framework of the biophysical model PARTRAC. The Escherichia coli catabolite gene activator protein (CAP) induces a bend of about 90 degree in its 30-bp sequence DNA binding site. The known three-dimensional date of the structure of the CAP-DNA complex was incorporated into the PARTRAC model of high-order DNA organisation. The Auger electron emitting iodine-125 atom was assumed to be located at a specific site near the center of the complex. From the interactions of the CAP-DNA complex with simulated track structures of iodine-125 decay, the direct physical hits and the indirect hits from OH radical attack are scored for the analysis of DNA strand breaks. The simulated distribution of DNA strand breaks is compared with available experimental results.
