COMBINING ATOMIC-FORCE-MICROSCOPY AND OPTICAL SINGLE-MOLECULE-MICROSCOPY — •O. Panzer, U.C. Fischer, and H. Fuchs — Physikalisches Institut, WWU Münster, Germany

In recent years various techniques for the detection of single molecule’s fluorescence have been developed. While first pictures of single fluorophores were obtained by aperture type SNOM [1] today conventional, more convenient microscopic methods are mostly used [2, 3]. On the other hand conventional optics cannot provide resolutions below the diffraction limit. We will present the combination of an AFM with a sample scanning confocal microscope, which is capable of detecting single fluorophores. We have put an AFM on top of the optical setup to localize single molecules with a resolution of about 10nm in future experiments. Therefore the AFM-tip will be used to perturb the fluorescence of single dye molecules. Fluorescence-quenching energy-transfer processes between the metal-coated tip and the molecules show typical ranges of less than 10nm [4]. Imaging the effect of these interactions would allow for the relative lateral localization of single fluorophores with a resolution of the order of 10nm.
