Insight into the dynamic structure of the cell nucleus by visualization of single protein molecules — •Ulrich Kubitscheck — Institut für Experimentelle Physik, Universität Bremen

The nuclei of mammalian cells are highly organized structures, in which chromosomes occupy defined territories, and complex supramolecular assemblies transcribe and replicate DNA in dedicated domains. These domains may often be visualized by fluorescence microscopy as intranuclear "specklesNo-dq of No-dqfociNo-dq. The distribution of splicing factors that process pre-messenger RNA is a prominent example for the high spatio-temporal organization in the cell nucleus. We have analyzed the structure and dynamics of nuclear supramolecular complexes such as the large pore complex in the nuclear envelope and also nuclear domains such as the speckles for the first time on the level of single protein molecules and RNA-particles. The visualization of single molecules within cell nuclei by highly sensitive and high-speed video microscopic methods allows detailed insights into the largely unknown dynamics of intranuclear structures.
