Photophysics of the Green Fluorescent Protein (GFP) on the single molecule level — •Gregor Jung and Andreas Zumbusch — Department Chemie, LMU München, Butenandtstr. 11, 81377 München
Since the discovery of the unique in-vivo labelling properties of GFP, this protein has attracted a lot of attention. The natural fluorescence is entirely due to internal interactions of aminoacids within the protein. Thus, the fluorophor acts as a probe of the local environment, the protein matrix. Mutations in the primary structure consequently lead to changes in both the static and dynamic fluorescence features of GFP. Information about the protein dynamics are obtained by studying fluorescence dynamics.
In our contribution, we show results of the past 5 year. The underlying investigations of 12 mutants have been performed using fluorescence correlation (FCS) and single molecule spectroscopy.
