Scanning confocal Raman microscopy gives the possibility to achieve material contrast of Raman-active samples in plane and in perpendicular direction due to the specific Raman vibration frequencies of different molecular groups. Three dimensional sample structures in the order of magnitude of a few 100nm can be visualized by Raman-contrast.
To examine the self assembly of biomaterials under static and dynamic confinement, microchannel diffusive mixing devices were used. The devices consist of two crossed microchannels (width 10-100µm) where in a continuous flow the diffusive mixing of biomaterials along the laminar stream profile can be observed. Using Raman-contrast imaging, the evolution of the interactions of the biomaterials can be evaluated by varying the scanning position along the reaction stream. Varying the velocity ratio of the incoming streams, we can control the reaction area and therefore we can tune the reaction velocity. In our first model system, we study DNA bundling induced by Co3+ ions.
