The interactions of immobilized single-stranded ARS121-1 DNA and replication protein A (RPA) were observed by surface plasmon resonance (SPR) using a BIACORE X. The analysis included the determination of the association rate constant ka, the dissociation rate constant kd and the equilibrium constant KD. From this the Gibbs free energy Δ G and the transition state free energy Δ G‡ were calculated. By measuring the reaction at different temperatures, it was possible to extract thermodynamic information like Δ H and Δ S about the system using van’t Hoff analysis.
The kinetic and thermodynamic information gained by SPR were compared with data observed by fluorescence correlation spectroscopy (FCS).
