The design of artificial cell membranes on solid surfaces, like glass or semiconductors, enables us to investigate the basic principles of complex functions in nature. Lipid membranes directly deposited onto solid supports cannot be used as a platform to incorporate large transmembrane cell receptors (e.g. integrin), since the separation between the membrane and substrate (1-2 nm) is not sufficient to keep the extracellular domains in a functional form.
In this study, we introduced well-defined hydrophilic polymer spacers to control the membrane-substrate distance. Careful optimization of the preparation conditions enabled us to fabricate homogeneous No-dqpolymer-tetheredNo-dq membranes, where the membrane-substrate distance and membrane fluidity can be precisely adjusted by the length and concentration of the polymer spacer.
