In bacterial photosynthesis photons are absorbed by light–harvesting complexes which pass the excitation energy efficiently to the photochemical reaction center (RC) where the excitation energy is used to reduce a quinone which finally leaves the RC. One of the light harvesting complexes (LH1) is closely asociated with the RC and it is assumed that it has a ring like structure that encloses the RC. However, such a supramolecular organization is questionable because it would prohibit the material transport required for the biological function of the photosynthetic unit. One conjecture is that another small membrane protein — PufX — may be part of LH1 and plays a key role in the regulation of the material exchange. In order to shed more light on the debate whether PufX is an integral part of the LH1 ring we perform a study that compares spectra from single wild type LH1–RC complexes with spectra from single PufX lacking mutants. The first results will be presented.
