Acidic lipids such as PIP2, PIP3 and PS are assumed to be key players in many signalling pathways. It has been shown in in vitro experiments that proteins with basic effector domains can bind to membranes containing acidic lipids and induce clustering of acidic lipids. One of those proteins is the cytosolic protein MARCKS (major alanine rich protein kinase C substrate), which is present in all cells of vertebrates. In quiescent cells MARCKS is membrane bound and colocalizes with acidic lipids. In response to various stimuli MARCKS undergoes a cycle of phosphorylation by a protein kinase C, subsequent detachment from the membrane, dephosphorylation in the cytosol and re-attachment to the membrane. We investigate theoretically the possibility that domains of acidic lipids are induced by MARCKS and discuss the consequences for signal transduction.
