The Orbitrap mass analyzer is an electrostatic trap that has been demonstrated to provide high resolving power, mass accuracy and high space charge capacity with pulsed ion sources. When interfaced to continuous ion sources however it proved to be more challenging when trying to combine these features with wide mass range, high sensitivity and dynamic range. This works describes a novel hybrid mass spectrometer where limitations of earlier schemes were successfully overcome. The entire instrument operates in LC/MS mode with nominal mass resolving power 60,000 (1 spectrum/sec) and maximum resolving power in excess of 100,000 (FWHM). Automatic gain control is used to provide user-controlled ion population thus ensuring high accuracy of mass measurements: <2 ppm using internal standards and <5 ppm with external calibration. More importantly, the instrument provides very high dynamic range of mass accuracy. Rapid and automated data-dependent MS/MS allows the instrument to operate in real time, with up to 3 MS/MS high-mass accuracy spectra per second. These analytical parameters are seamlessly combined with MSn capability of the LTQ mass spectrometer. Examples of applications include analysis of pharmaceutical and proteomic samples using LC/MS(n) and demonstrate high sensitivity and specificity of the instrument.
