3D micro-spines for cells spatial guiding on Multi Electrode Array (MEA) — •Francesca Santoro, Gregor Panaitov, and Andreas Offenhäusser — ICS/PGI-8 Bioelectronics, Forschungszentrum Jülich GmbH, Jülich, Germany

For analysis of recordings of neuronal activity it is very important to study samples with well defined interconnections within neuronal networks. This kind of neuronal systems can be developed by guiding of individual neuron cells during the growth. In a rat cortical cell culture, we applied 3D gold micro-spine arrays for spatial guiding of dendrites growth on a defined geometrical pattern. We report on results about fabrication of micron and submicron gold spines by electron beam lithography and electroplating. Furthermore, prior to cell culturing the gold surface was functionalized by Amino-Poly(ethylene glycol)-thiol Self Assembled Monolayer (SAM). Well-defined guidance of neurons by artificial micro-spines array was demonstrated and a detailed investigation with Fluorescence Light Microscopy and Scanning Electron Microscopy techniques was carried out. Moreover, the fabrication of micro-spines onto MEA-electrodes (Multi Electrode Array) demonstrated improvement of contact interface between the electrode and cell membrane, which is important for recordings of neurons action potentials.
