The interaction of proteins with the intracellular side of cell membranes is important for a variety of cellular functions. We are mainly interested in cell adhesion and migration and therefore, in the interaction in between integrins, proteins of the integrin complex and f-actin.
To study this interaction, we extract the cell membrane with the unroofing technique [1] and covalently crosslink substrate with cell membrane in vitro to stabilize the system. The cell membranes of adherent cells are prepared in such way, that the intracellular side is exposed.
Furthermore, we verify our sample preparation with the help of atomic force microscopy, fluorescence light microscopy and scanning electron microscopy.
