Our group is investigating the differences in cell morphology and physical properties of head and neck squamous cell carcinoma cells (HNSCC) and oral keratinocytes. As the cytoskeleton plays an important role in elasticity and mechanical properties of the cell, it is a significant part of our research. The cell lines, originating from different locations of the oral mucosa are investigated using a confocal laser scanning fluorescence microscope. To obtain more information about the cell morphology with a focus on the cytoskeleton, the cells were stained using SiR-Actin (actin filaments) and SiR-Tubulin (microtubules) staining kits. Three-dimensional images of the cells were used to compare the cytoskeleton of the different cell lines in size, volume and shape. The cytoskeleton of cell lines originating from the same area show similar properties whereas cell lines from other head and neck areas show differences. Our latest results will be presented and discussed.
