Skeletal muscle makes up the majority of human muscle tissue. Its contraction allows for controlled movements of the body, as well as being essential for maintaining posture. To understand the global and local forces at play during such contractions, occurring on the time scale of seconds, a 3D high-speed high-resolution imaging method is necessary. This can be achieved with a multiview light-sheet microscopy setup, providing isotropic resolution of the sample. We present a new FEP-based culture chamber design for raising biomimetic muscle tissue anchored between two mm-sized post, enabling unconstrained image acquisition from any rotational angle. Muscle contraction can be triggered by electric pulses through chamber wiring.
