For several years a number of techniques made it possible to detect the fluorescence of single molecules. The characteristics of many fluorophores were explored since, they were utilized as probes of their nano-environment and phenomena specific to single molecules were observed for the first time.
Very recently a variety of experiments have been proposed to manipulate the optical properties of single fluorophores in a controlled manner, mostly by energy transfer to a functionalised probe.
To enhance the spatial localisation of single molecule fluorescence microscopy we want to make use of short ranged fluorescence-quenching in the near-field of the metalized tip of an atomic force microscope (afm). Therefore we combined a scanning confocal fluorescence microscope with a tuning fork type afm. Here we present first results concerning the apparatus and samples.
