GroEL is a protein complex with a toroidal shape and a central cavity. GroEL, a chaperonin molecule, folds misfolded or newly synthetized proteins. The activity cycle of GroEL implies a change of the central cavity from hydrophobic to hydrophilic character. By means of the Atomic Force Microscopy it was possible to show at the single molecule level that the central cavity changes from hydrophobic to hydrophilic during the ATP cycle. This measurement was carried out by exploiting the attractive or repulsive interaction between the GroEL cavity and the AFM tip.
