The fluorescence of endogeneous fluorophores as NADH, FAD, and Protoporphyrin IX is a useful tool in research and clinical diagnostics as these molecules act as markers for metabolism activity. In physiological samples unfortunately the strong autofluorescence from connective tissue, namely Collagen and Elastin, becomes a problem under UV illumination.
We can show how this problem can be overcome by two photon excitation with fs-laserpulses in the near infrared. Under this excitation conditions the fluorescence behaviour of Collagen and Elastin drastically changes while the aimed Fluorophores act as under single photon excitation.
